The effects on the proteome of increased levels of endogenous CKs in dark grown Arabidopsis seedlings were studied. Increase in endogenous CK levels was achieved by inducing the CK biosynthetic gene, ipt. Image and mass spectrometric analysis of proteins separated in two dimensional gels identified a number of differentially expressed proteins, most of which are related to chloroplast biogenesis and function. All the identified chloroplast proteins were upregulated in response to increased CK levels. In contrast, all differentially regulated mitochondrial proteins, representing the second largest protein group, were downregulated. Comparison of our proteomic data with previously published transcriptome profiling of light induced photomorphogenesis and effects of increased CK levels in light grown Arabidopsis seedlings revealed the same general trends for most of the 23 transcripts corresponding to the proteins in our data set.
