Description | Specificity and sensitivity of polyclonal antibodies prepared after immunization of rabbits with antigens (extracted from mycelial mats of Phytophthora nicotianae var. nicotianae-PNN) were studied. Antisera and IgG were suitable to detect other Phytophthora spp., i.e. P. cinnamomi, P. cactorum, P. cryptogea, P. fragariae; differences between the species were quantitative. The specificity of polyclonal antibodies was increased with saturation of IgG with antigens. There was no increase of polyclonal antibodies specificity and sensitivity after immunization of rabbits with purified PNN antigen or fraction of proteins. Nor was there a cross-reaction of anti-PNN IgG with antigens of Fusarium oxysporum, Pythium oligandrum, Pythium ultimum and Clavibacter michiganensis subsp. michiganensis (tomato isolates). Reactions of antisera and IgG with antigens were evaluated by agar double diffusion and ELISA. |
