Excreted/secreted products of entomopathogenic nematodes and their effect on insect immunity

Popis Entomopathogenic nematodes (EPNs) are the obligate parasites of insects. In the fight against the immune system of the host, nematodes use their excreted/secreted products (ESPs). These products are a mixture of molecules with distinct functions. We identified several functions of the ESPs and examined them in detail - specifically their influence on phenoloxidase activity (PO) and coagulation. ESPs were obtained from two isolates of Heterorhabditis bacteriophora from different environments. Firstly, we purified the samples based on their charge. There were 5 fractions obtained from each isolate of H. b. in which their activity in the inhibition of G. mellonella PO was verified. Inhibition activity was also observed for the coagulation in both isolates. For the active fractions in PO inhibition were used a second purification based on their size. We obtained 18 new fractions in which we found very highly activity which was manifested in the inhibition of phenoloxidase and coagulation. Phenoloxidase and coagulation inhibition were also tested on ESPs isolated from EPN Steinernema carpocapsae. The inhibition of both immune responses was confirmed. Potential use of these results is mainly in the biological control of insect pests.
