The effect of single-species Dactylogyrus infection on the expression of immune genes in gibel carp (Carassius auratus gibelio)

Popis The immune response varies during parasite infection. However, the studies concerning sin-gle-species monogenean infection and genetic variability of immune genes in fish are scarce. In this study, the effect of single-species infection, i.e. the infection with Dactylogyrus falci-formis (Monogenea), on the expression of selected immune genes in gibel carp (Carassius auratus gibelio) was investigated. To ensure the single-species infection, adult specimens of Dactylogyrus species were collected from fish sampled in Dyje River and were kept individu-ally in the wells with water until the eggs were laid. All parasites with eggs were subsequently determined under the microscope, and only eggs from Dactylogyrus falciformis were used in the following experimental study. Hatched oncomiracidia were used for infection of triploid specimens of gibel carp. Fifteen parasite-free fish divided into five groups (each represented by three specimens) including one control group and four experimental groups were infected by 0, 5, 10, 20 and 50 oncomiracidia, respectively. Fish were dissected 21 days post-infection, gills were checked for parasites, the selected organs (spleen, thymus, head kidney and intes-tine) were sampled and used for molecular analysis of immune genes expression. RNA-sequencing of spleen samples and subsequent bioinformatic analysis of obtained data were used to determine all differentially expressed genes. Afterwards, based on data from RNA-sequencing and literature review, genes with immune-related function (HMGB5, SOCS1, TGF1, TNF1 and STAT5) were selected for further analysis. The expression of these genes in the spleen, thymus, head kidney and intestine was later quantified using RT-qPCR. Differen-tial expression of selected genes between experimental groups and between immune organs was observed and statistically evaluated. This study presents the evidence of differential ex-pression of five genes in lymphatic organs of gibel carp during single-species infection by Dactylogyrus falciformis. It provides a better understanding of this host-parasite system and may offer a general outline of immune genes expression in response to infection by Dactylo-gyrus species.
