Popis | Resistance to tyrosine kinase inhibitors (TKIs) is a major issue in cancer therapy, as almost every patient undergoing this treatment gains chemoresistance. In this project, we attempted to develop chemoresistant cell lines insensitive to three recently approved TKIs - vandetanib, lenvatinib and cabozantinib. Our goal was to obtain these cell lines by continuous growth in the presence of said inhibitors for further examination and research of chemoresistance mechanisms. Unfortunately, we came across some problems and the long-term character of our experiment forced us to take up a different approach. In the future, we plan to continue our project by inducing chemoresistance with transfection of an expression vector containing mutated sequence of receptor tyrosine kinase. |
