Popis | Ultrastructure of Cryptococcus neoformans was studied to characterise morphologically different stages of the cell division cycle. Six stages were distinguished: 1) A single mother cell with a filamentous capsule on the multilamellar cell wall and typical membrane organelles (nucleus, mitochondria, vacuoles, endoplasmic reticulum and secretory vesicles). 2) The cell with a cylindrical protrusion - stalk. 3) The cell with an isodiametrical bud differentiated at the end of the cylindrical stalk. 4) The cell with a nucleus that migrated into the bud and subsequently entered mitosis. 5) The cell after nuclear division, but before cytokinesis. 6) The cell at cytokinesis forming septum in the bud neck and separation of the daughter from the mother cell. Freeze-substitution provided a better resolution of intracellular components, while the outer structures - wall and capsule were better seen after glutaraldehyde fixation. |
