DNA is central to multiple key biological processes, such as transcription, DNA repair, and DNA replication. Only recently it has become evident that transcription is a major source of DNA damage, and that RECQL5β is central in preventing transcription-borne DNA damage. We hypothesise that RECQL5β acts via suppressing RNA polymerase II (RNAPII) - mediated transcription. Currently, only limited information, concerning the detailed molecular mechanism of action of RECQL5β is available. Recent advancements in cryo-electron microscopy have enable us to address the role or RECQL5β in suppressing transcription-borne DNA damage from a structural point of view. We will therefore determine 3D structure of complex between mammalian RNAPII and RECQL5β—a first such structure to the date. We expect to uncover biochemical and molecular mechanisms underlying the suppression of transcription-borne DNA damage by RECQL5β, which will serve as a proxy in understanding the cross-talk between transcription and DNA repair.
