Melanoma is commonly driven by oncogenic mutations over-activating the ERK signalling pathway. We found that metabolic stress can affect ERK activity in melanoma, through the AMPK kinase. Crucially, we observed decreased AMPK activation in cells with a disrupted lysosomal Ragulator complex. This project aims to determine the role of Ragulator in the crosstalk between ERK and nutrient-sensing pathways AMPK and mTORC1 in melanoma. Our findings could lead to new therapeutic strategies for melanoma.
