In vitro and In-cell characterization of Quadruplex-duplex hybrids: conformation, folding, ligand binding and implications for in-cell NMR based assays (QDHassay)

The research program involves (1) developing 19F NMR probes for characterizing G4 folding in the presence of a complementary C-strand and validated by ESI-MS. (2) Elucidating the complex G-quadruplex folding pathways from different G4-duplex, G4-duplex-G4, duplex-G4-duplex system by combining for the first time mass spectrometry, nuclear magnetic resonance, (3) In cell 19F NMR to evaluate the impact of ligands in different phases of cell cycle and compare with in vitro data. Our project will shed light on the biologically relevant picture of the dynamics of G4 structures at the molecular level. It will also foster collaboration between European researchers specialized in different biophysical and structural approaches to study G-quadruplexes. Finally, it will also contribute to society by improving analytical measurement approaches highly relevant to pharmacology.
