Cytokinins (CKs) are known to act in combination with other phytohormones to regulate diverse developmental processes in plants. However, knowledge of molecualr mechanism of CK action is limited. We will use a novel transcription activation system for regulated gene expression to create transgenic plants overproducing individually heterologous enzymes generating free CKs either by glucoside hydrolysis or by de novo synthesis in defined spatial and temporal patterns. We expect that CK release from glucosides will cause. in addition to common effects. several distrinct effects compared to de novo CK synthesis. Analysis of the distinct effects will contribute to understanding of complexity of CK metobolic conversions. This approach will lead to isolation of Arabidopsis lines displaying phenotypes corresponding to defined CK overproduction patterns. In future, the lines will be used, via isolation of suppresor mutations, to isolate genes involved in CK metabolism, and CK signal pathways.
