Nuclear topology of some protooncogenes in human neutrophilic granulocytes and leukemic cells

The project will study the topology of C-MYC, ABL and BCR protoocogenes in the nuclei of human granulocytes in comparison with the topology of mentioned genes in leukemic cells that will be differentiated along the myeloid pathway taht is characteristic by changes of gene expression. The relationship between the structure of nuclear chromatin and gene expression will be studied not only in the model differentiation experiments, but also in the parients suffering from chronic myelogenous leukemia (CML). The differences in the topology of ABL and BCR genes will be detected at the time of chronic, accelerated phases and at the time of blastic crisis of CML which differ in the expression of ABL gene that is translocated into the region of BCR locus (Philadelphia chromosome). Amplification of the C-MYC gene will be also assessed. The aim of this work is to find the differences in the topology of some genes that will be detected in the CML patients in contrast with the healthy individuals.
