Ischemic brain injury (stroke) represent a major socio-economic burden, with one in four people affected over their lifetime. Despite tremendous effort invested into development of new neuroprotective strategies, early clog removal remains the only approved therapy. The lack of new therapies is caused by the complexity of the disease, the dominance of neuron-centric research, the complicated drug design, and the application of inappropriate experimental models. Here, we propose to tackle these challenges with multidisciplinary approach, involving advanced stroke modelling in organoids, deep molecular understanding of the pathophysiological processes by single-cell transcriptomics, focus on non-neuronal cells, and effective mRNA-based therapeutic intervention. The combination of the advanced technologies and the experience of team members will lead to establishment of a unique platform for modeling of stroke pathophysiology and mRNA-based intervention, which will extent the current spectrum of stroke models in pre-clinical research.
