Description | A fucose specific Aspergillus fumigatus lectin, AFL1, was subjected to amino acid selective labelling by 15N. The the assignment of key amino acids for the binding can be readily achieved by use of single point mutants. Here we present optimization of recombinant AFL1 lectin over-expression in E.coli cells which is performed in 15N Trp selective supplemented M9 minimal medium. (Semi) optimized parameters were E.coli strain type, concentration of antibiotic, concentration of IPTG for induction, temperature after induction and length of lectin expression after induction. |
