Description | Gold nanoparticles (GNPs) represent a very attractive material for the construction of sensors and biosensors with immobilized aptamer on the surface, due to their biocompatibility. The most extensively studied aptamer is the thrombin binding aptamer (TBA) with sequence GGT TGG TGT GGT TGG, specifically binding to thrombin, and thus inhibits the activity of thrombin in the cascade of reactions resulting in the hemocoagulation. The aim of our contribution is spectroscopic characterization of aptasensor system and verify its specificity to thrombin in buffered and non – buffered aqueous solutions with adjusted ionic strength. For our experiment the GNPs (5, 10 and 20nm) stabilized in citrate buffer were used. The absorption spectra were monitored not only in the dependence on particles size, but also on the components of used solutions. For the detection of aggregation and behavior of unmodified and modified GNPs the distribution of GNPs and their zeta potential were determined. |
