Description Inducible genes controlled by signalling cascades may be regulated as continuously variable or as two-state on/off switche. We describe a novel stochastic model of the interface between signal transduction and transcription initiation that accommodates both possibilities. The main challenge has been the need to reconcile the dynamics of a single copy of each gene interacting with a large population of signalling molecules. Comparison of measurements of mRNA levels in live cells with the outputs of the model allows us to test its conformance with experimental data. Our approach dissects the characteristics of the interface, an essential mechanism driving the initiation as well as the production of mRNA. The model provides a range of predictions for initiation and mRNA profiles as a function of the interface parameters, and may now be more closely aligned with further experimental observations.
