Description | Lyme disease is multi system bacterial disorder caused by B. burgdorferi sensu lato. This spirochete is characteristic by being rich in antigenic determinants enabling adaptation to broad spectra of hosts. One of the most important antigens is a lipoprotein OspC essential in transmission to vertebrate host. The aim of our work what to prepare recombinant forms of OspC relevant for the region of the Czech Republic and isolation of monoclonal antibodies against these polypeptides. The antigen used in immunization experiments was derived form strains isolated at Brno-Pisárky locality (Czech Republic). The most abundant OspC type G4 was chosen for immunization. Nine different hybridoms were selected based on ability to bind native as well as recombinant OspC in solution (ELISA) and/or in denatured form (Western-blot). For three isotypes of monoclonal antibodies the recognized epitope was determined. |
