Description | The aim of the contribution is the comparation of selected methods of water samples analysis for sensitive isolation and subsequent detection of viral particles. The methods of filtration through glass wool columns, membrane filters and direct organic flocculation were tested as isolation methods. In order to verify the effectiveness of the selected methods and to perform their optimization, artificially prepared viral particles derived from bacteriophage MS2 with a unique insert sequence were used. The reverse transcription polymerase chain reaction was used for detection. This method allowed quantification of viral load in sample and thus determination of the effectiveness of the entire analytical procedure. Based on the results and experience, the direct organic flocculation method was chosen as the most appropriate method for isolation of viral particles from water samples. Compared to other methods, it was repeatedly possible to achieve comparable efficacy values . The method was also advantageous because of its simple and time-consuming protocol. |
