Description | Znojmo-Hradiste is a Great Moravian location near the town of Znojmo. From the discovery in 2007 to 2015, more than 500 objects were discovered, most of which were burial pits. The research of this unique site is multidisciplinary and includes a variety of disciplines such as anthropology, genetics, and reconstruction of the appearance. Determining the visible phenotypic characteristics of DNA is a part of efforts to reconstruct the look of a person of unknown appearance. The human genome contains 6 genes that influence the determination of eye color. The most influential is the rs12913832 polymorphism of the HERC2 gene. Two methods were used to detect the rs12913832 polymorphism. 20 samples of DNA from Znojmo-Hradiste were analyzed using a pre-designed TaqMan® probe, the marker was detected and the phenotype was estimated for 19 samples. The genotype was determined as a T / C assay for 15 individuals, 4 subjects were determined to be homozygous with the CC alleles in the detected locus. The second detection method was based on the Irisplex system. Primers from this system were used for conventional PCR and Sanger sequencing. A total of 12 aDNA samples from the Znojmo-Hradiste burial site were analyzed, of which 3 were able to determine the genotype of all six polymorphisms. For another 5 samples the rs12913832 polymorphism of the HERC2 gene was detected. The final sequencing results correlated with the TaqMan® assay results. |
