Description | Capillary electrophoresis (CE) technique is attractive for its rapid analysis, high efficiency, possible automation and nearness to physiological conditions. Despite of mentioned advantages of CE there is certain limitation of the technique, i.e. the low concentration sensitivity associated with the conventional ultraviolet visible (UV-VIS) detection. There are possibilities of increase the concentration sensititvity. Besides of exchanging the detection device with a more sensitive one, there are more elegant methods such as preconcentration techniques that allow the use of universal and less expensive UV-VIS detection. On-line preconcentration techniques allow automated analysis during separation while analytes are concentrated in the capillary [1]. As part of the miniaturization process for pharmacokinteic studies of plasma proteins-drug interactions, new more sensitive methods based on combination of the CE method with on line preconcentration techniques were optimized in this study. The established CE mode for affinity interaction studies is capillary electrophoresis-frontal analysis (CE-FA). The aim of this study was to optimize the new more sensitive CE-FA method in combination with on line preconcentration technique and the classical CE-FA method with similar conditions for data comparison. Another task was to consider evaluation of the binding properties of the drug plasma protein interaction by newly optimized method. |
