Description | The study was undertaken to characterize changes of the activities of oxidative and conjugative hepatic enzymes, CYP2D1 and NAT2, induced by pretreatment with fluoxetine or paroxetine. Method of the isolated perfused liver from the male Wistar rat donors pretreated in vivo with SSRIs was used for the study. The results showed that the metabolic capacity of the liver for O-demethylation of dextromethorphane to dextrorphan was significantly decreased. Pretreatment with fluoxetine, but not with paroxetine, evoked also inhibition of activity of NAT2. Thus, SSRIs showed significant inhibitory effects upon the enzyme activities in the rat liver. |
