Description | Plasmid pBR322 DNA was purified by gel permeation chromatography. Recovered plasmid DNA was free of RNA and retained its biological activity. The biological activity of purified plasmid DNA was tested by electrotransformation into bacterial cells. DNA fragments of lambda DNA and lambda DNA were separated on column for gel permeation chromatography. Small fragments were eluted in void volume; large fragments and lambda DNA were separated in reversed order than expected according to GPC and according to the slalom chromatography mechanisms. |
