Description | BM2 cell line was prepared by infection of chicken myeloid progenitor cells with AMV transducing v-myb oncogene. resulting monoblasts constituively express v-myb and their terminal differentiation steps are therefore blocked. BM2 differentiation pathway can be resumed using TPA, trichostatin A or by ligand-activated retinoic acid receptors. In this work we focused on characterization of of differentiating BM2 cells and search for protein markers useful for differentiation studies. We showed that dominant protein of 55 kDa appears in nuclear fraction of TPA-treated BM2 cells. Interestingly,this protein is absent from BM2 cells expressing human RAR and treated with TPA/RA. this protein was analyzed by SDS-PAGE, extracted from the gel and molecular mass of resulting peptides was determined by mass spectrometry MALDI-TOF. We compared our data with mass spectra in databases in identified this protein as vimentin. We confirmed rhis conslusion experimentally using vimentin-specific antibody. Because vimentin-expression does increase in BM2 cells induced by TPA or trichostatin A and does not increase in BM2RAR cells treated with RA we conclude that vimentin is a suitable marker for distinguishing of BM2 differentiation pahways |
