Description Magnetic microbeads carrying specific ligands can be used for capturing and separation of target cells or DNA from the environment containing extracellular PCR inhibitors or competitive microflora. The presence of magnetic particles itself or the individual components used in their preparation can decrease PCR sensitivity or lead to false negative PCR results, too. Thus a method which enables evaluation of magnetic particles non interfering in PCR was developed. The effect of magnetic particles and components on PCR sensitivity was tested using DNA isolated from Bifidobacterium longum cells. Newly prapared magnetic carriers (8 based on ferrites and 7 based on methacrylate polymer particles) were tested. The particles selected were used in PCR and RAPD applications.
