Description Sixteen hybrid U/Y proteins were constructed. Three amino acid residues of colicin U and four amino acid residues of colicin Y were found to be important for interaction with cognate immunity proteins. The amino acid residues F576, G580 and L594 were found to be involved in the interaction between colicin U and its immunity protein. Another interaction region is located in the position 590-592. The amino acid residues Y586, V590, A594 and A595 were identified in recognition of colicin Y by its cognate immunity protein. In addition, one of the 34 amino acids in colicin Y C-terminal region (12 amino acids are different when compared to sequence of colicin U) had a slight effect in the recognition of colicin Y with its immunity protein. Important amino acid residues of colicin U and Y interacting with cognate immunity proteins were found to be located in the hydrophobic sequence between helices 8 and 9 in the colicin pore-forming domain.
