Description In our experiments, we studied growth and development of early somatic embryos (ESEs) cultures of Norway spruce (Picea abies). The growth of ESEs is influenced by sub-cultivation. It was ascertained by image analyse method that increase of SRSE area is dependent on initial size of sub-cultivated part of original ESEs and on spatial orientation of embryos. In the concrete area of ESEs less increased if the spatial orientation was not kept original. Orientation influence on ESEs growth increased together with the age of culture. On the basis of the mentioned results, embryos viability in individual ESEs parts was determined by double staining using fluorescein diacetate and propidium iodide. Embryos viability was higher in outer parts of the cluster then in inner parts of cluster. The embryos obtained from different cluster parts were sub-cultivated and their subsequent growth was monitored. Increase of ESEs clusters area sub-cultivated from outer cluster parts was higher in comparison with area increase of ESEs clusters from inner cluster parts. That is why differentiation of Norway spruce early somatic embryos culture was confirmed.
