Description In our study, we have determined dioxin-like toxicity of 32 soil samples collected in two localities with respect to possible existing or prospective traffic contamination. All samples have been collected along the existing road between Ruzyně and Březiněves in Central Bohemia. 16 samples were collected in distances approximately 1m and 20m (8 samples each) from the existing road, the other 16 (8 samples of forest soil and 8 samples of arable soil) along a highway to be built. In vitro measurements of the dioxin-like activity of crude extracts were carried out using H4IIE.luc cell line (a cell line producing luciferase under transcriptional control of Ah-receptor), and the results were compared with data obtained by chemical analysis of PCBs and PAHs. There was a good correlation between toxic equivalents (TEQs) computed from PAHs concentrations and TEQs obtained from the in vitro assay. We have observed a significant increase in concentrations of PAHs and dioxin-like activity in samples taken close to the road when compared to those in 20m distance. Soil sampled in 20m distance from the road (arable soil) showed no or only moderate increase in dioxin-like toxicity and PAHs concentrations in comparison to soil in unpolluted area (where the road is yet to be built). An interesting result is a significant difference between dioxin-like activities of arable and forest soil in the clean area. Samples of forest soil elicited significantly higher dioxin-like activity than arable soil. This may be caused by moving pollutants into deeper layers of the ground during ploughing of the arable soil, or by higher content of organic matter in forest soil and thus to slower shift of pollutants into the deeper layers.
