Description | Restriction mapping was used for comparison of four treponemal genomes including strains T. pallidum subsp. pallidum Nichols, T. pallidum subsp. pallidum SS14, T. pallidum subsp. pertenue Samoan D and T. paraluiscuniculi. The different degree of invasivity and pathogenicity of these strains could reflect the differences in their genomes. The genomes of analysed strains were divided into 94 intervals (TPI intervals) covering the whole genome and each of these segments was amplified by XL PCR. These PCR products were digested by restriction enzymes BamH I, EcoR I and Hind III and the resulting restiction profiles were used for identification of heterologous regions in chromosomal DNA. The genome differences are preferentially localized in tpr genes. Identified nucleotide changes can be used for molecular identification of individual treponemal strains or isolates. |
