Description | Differences in the cell-specific expression of intermediate filament proteins in neoplastic tissue represent an important tool for tumor diagnostic. According to published studies, the re-expression of nestin has been detected in tumors derived from CNS tissues, as well as in some other types of human solid tumors. It is evident that nestin re-expression in tumor cells is closely related both to the dedifferentiation and malignancy of the respective tumors. Therefore, immunohistochemical and/or immunocytochemical examination of nestin may serve as a useful tool for classification and accurate grading of human malignancies. Our study was focused on a detection of nestin in cell lines derived from several types of human solid tumors. By employing indirect immunofluorescence, we proved a strong re-expression of nestin in NB-01 (neuroblastoma), JE-03 (malignant fibrous histiocytoma), JE-04 (osteoplastic osteosarcoma) and GM-7 (glioblastoma multiforme) cell lines. A detailed study of the nestin expression both on intracellular and ultrastructural levels was performed using GM-7 cell line; the results shown the presence of nestin not only in the cytoplasm but also in the cell nucleus. |
