Description | Mutations of the p53 gene have been found in over half all human cancers. There are several techniques available for detection of p53 mutaations including FASAY (functional analysis of separated alleles in yeast). We analyzed the p53 status in various human tumors by FASAY, including acute myeloid leukemia, myelodysplatic syndrome, malignant melanoma, head and neck squamous cell carcinoma, breast and ovarian cancer, leimyosarcoma and chronic lymphoid leukemia. We identified 22 temperature-sensitive p53 mutations and localized most of them to the part of the gene coding for DNA-binding domain. Functional outomes of individual mutations were addressed. Deatailoed analysis and stratification fo p53 mutants coubd be helpful for better understanding of the effects of specific turmorogenic mutations and for therapy designed to restore function of the p53 protein. |
