Cytotoxic and colour properties of quaternary benzo[c]phenanthridine alkaloids

Description | Our results show that IC50 values (0,03-3,7 microg/ml) after 72 hours treatment of alkaloids are comparable with IC50 of routinely used cytostatics. Cytotoxicity of individual tested alkaloids descended in this order: MA ; SL ; CHL ; SR ; CHR ; SA ;CHE. HL-60 and A-2780 were the most sensitive cell lines while HeLa cells were the most resistant. All tested alkaloids appeared to be inductors of apoptosis at the concentration 1 microg/ml. In addition, we have observed interesting fluorescent characteristics of some QBA upon intercalating into DNA of living cells. We have proved that SA, MA, CHR and partly SR, can be used as supravital fluorescent DNA probes both in fluorescence microscopy and flow cytometry. |
