Description Gel electrophoresis (GE), a selective component, is combined effectively with LA-ICP-MS as a sensitive component, and with other mass spectrometric techniques as structural components of metalloprotein associated approach. Native and denaturating GE was done for model sample of BSA saturated with Cu(II+), Co(II+) ions. Sample was preconcetrated for 10 mins at 100 V, separation took 30 mins at 200 V. Gel was then dried in gel drier (model GD5040). A nanosecond Nd:YAG laser (New Wave, USA) working at 213 nm with pulse length 4.2 ns and frequency 20 Hz was used for laser ablation of separated metalloproteins. Dried gel sample was in 10cm-3 ablation cell. Lowest RSD values for signal intensity were obtained with gels stained with silver. During laser ablation optimisation of gel saturated with Co(II+) ions it was found that optimal densities of irradiation output lie between 0.2 GW.cm-2 and 2 GW.cm-2.
