THE INFLUENCE OF POLYPHENOLIC EXTRACTS FROM GRAPE BYPRODUCTS ON THE METABOLIC ACTIVITY OF CYP1A2 IN HYPERCHOLESTEROLAEMIC RATS

Description | The enzymatic system of cytochrome P450 (CYP450)is one of the major metabolic enzyme systems. It takes part in metabolism of many exogenous as well as endogenous substrates and its activity may be influenced by many factors (age, sex, pathophysiological status). One of these factors which is often missed out is diet. Food contain many substances influencing activity of CYP450 and thus biotransformation of potentially toxic agents including drugs. The aim of our work was to prepare extracts from grape byproducts rich in polyphenols and to investigate their influence on the activity of hepatic CYP1A2 in hypercholesterolaemic rats. Phenacetine (PHEN) and paracetamol (PAR)were used as marker substance and specific metabolite in the model of isolated perfused rat liver. Extracts were a administered to male Wistar albino rats by intragastric sond for 40 days. Rats were suplemented by hypercholesteroleamic diet (1,7%). Hypercholesterolemic diet alone had no effect on the activity of CYP450. Induction of the activity of 1A2 isoenzyme evoked by extracts administration manifested as increase of PAR levels in perfusate when compare to control animals. We have also observed atypical increase of PHEN levels in the beginning of the perfusion in animals treated with extracts. |
