Description | Backgrounds: Early diagnosis and immediate therapeutic interventions are crucial factors for successful cancer treatment. New biomarkers are needed for even earlier diagnosis, more precise histopatological classification or for prediction of treatment response. Our study was aimed at the differential analysis of protein expression profiles of cancer and non-cancer tissue from 105 patients with breast cancer. Methods and Results: Tissue lysates were analysed by SELDI-TOF MS on copper chelating chips IMAC 30. Peak intensities were statistically evaluated by Wilcoxon test with p<0,01. 28 peaks are over expressed in tumour tissue and 18 peaks are over expressed in normal tissue. Protein with molecular weight 15841 Da was identified as beta chain of haemoglobin by method peptide mass fingerprint (PMF). Conclusions: Analysis of protein expression profile by SELDI-TOF MS is able to provide a new point of view on diagnostic and tumour classification. Further independent studies of new sets of samples are essential for verification of specificity and sensitivity of published potential biomarkers. |
