Description | SELDI-TOF MS is a protein profiling method often used in clinical proteomics for screening and biomarker discovery. Our effort in last year has been focused on identification of proteins of interest in projects based on SELDI analyses of rat plasma and breast tissue using high resolution mass spectrometry. Contributions of a panel of protein separation and/or identification method like tricine SDS-PAGE, 2-D PAGE, LC-MS/MS, FT-ICR, iTRAQ-2DLC-MS/MS for the biomarker identification in SELDI studies will be presented. As a result of these experiences, we have developed a method combining IMAC column preseparation, reverse phase LC, SELDI protein profiling, tricine SDS-PAGE, protein profile matching and MS/MS identification. Using this approach, we have identified several proteins potentially related to breast cancer development and cytostatic agens action. |
