Description | We found that chemical transfection with Fugene HD and a Tet-On 3G Inducible Expression system, EF1a Version (Clontech) are the methods of choice for derivation of inducibly everexpressing clones of human ES cells. Our data also suggest that combined PCR based linear DNA fragments are superior over plasmid based transfection for establishing bioengineered sublines of hES cells inducibly everexpressing complex transgenes. |
