Characterization of novel molecular players involved in auxin regulated protein trafficking in plants.

Regulation of growth, development and plant adaptational capacity depends largely on generation and maintenance of auxin gradients. The distinct spatial distribution pattern of this hormone in plant tissues is achieved by its directional flow accomplished by polarly localized PIN efflux carriers.
Notably, auxin influence the localization of PIN proteins, by inhibiting endocytosis and promoting their retention on the plasma membrane and thus increasing the efflux of the hormone itself. Although, recent studies indicate auxin regulating early steps of internalization and endosomal trafficking, the mechanistic understanding of this process is still lacking. To fill this knowledge gap we conducted a forward genetics screen in which we identified novel potential regulators of this phenomenon.
Those candidates exhibit defects in auxin-mediated inhibition of endocytosis showing PIN1-GFP internalization in presence of auxin. We aim to perform a characterisation of those candidates that will prospectively contribute to unravelling of the developmentally vital auxin feedback mechanism.
