Identification of cytokinin receptors of Brassica napus

Popis | Cytokinins (CK) belong to a group of plant hormones, playing crucial role in plant growth and development. CK signal is transduced through a multistep phosphorelay where the phosphoryl group is transferred between His and Asp residues. Cytokinin perception has been well-studied and the CHASE-containing sensor His kinases (CHKs) have been shown to be important morphological factors in Arabidopsis. As Brassica napus shares a high degree of nucleotide identity in coding regions with Arabidopsis, we used sequences of A. thaliana His kinase genes (AHKs) to search for their orthologs in B. napus (BnCHKs). Experimental screen of bacterial artificial chromosome (BAC) genomic DNA library of B. napus, var. Tapidor (JBnB), resulted in identification of redundant number of overlapping BAC clones, which hybridized with PCR-generated probes derived from sequences of phylogenetically conserved AHK regions. Five selected clones have been sequenced and defined as CK receptors by computational analysis. Based on prediction, gene-specific primers have been designed and coding- as well as untranslated cDNA regions cloned. Gene structure analysis and prediction of conserved structural domains of five BnCHK proteins are presented. Four homologues of AHK2 and one homologue of AHK3 have been identified by this experimental approach. |
