Popis | During the period 2008-2012, we performed sampling of both allopatric as well as sympatric barbel population along the hybrid zone of these two species. The aim of our project is first to study genetic diversity of barbel populations with the use of molecular markers and to investigate the composition of metazoan parasite communities in both allopatric and sympatric populations. MHC class IIB genes, the perfect candidates to study host-parasite interaction at the molecular level, will be studied further to test hypotheses of co-adaptation between immune system and specific parasites. Finally, association between host specificity and the degree of gene introgression along the hybrid zone will be analyzed. So far, 30 microsatellite markers were isolated and pooled into the five multiplex sets. On the basis of Bayesian analysis performed in the program STRUCTURE, de novo developed microsatellites showed to be the perfect tools to differ between hybrids and pure parental species (Gettová et al., unpublished). |
