DNA-based identification and OspC serotyping in cultures of Borrelia burgdorferi s.l. isolated from ticks collected in the Moravia (Czech Republic)

Popis | In Europe nine different genospecies responsible for the human form of the LD have been reported. Among those B. afzelii, B. garinii and B. burdgorferii s. s. are the most prevalent in the Czech Republic. Each genospecies can be further subdivided into specific genotypes depending on genetic marker used in analysis. In our study we used two different loci (flaB and ospC) to assign genospecies and OspC phylogenetic type to eighteen strains previously isolated from ticks collected at a locality Pisárky, a suburban park near the city of Brno (Czech Republic). The RFLP analysis revealed three different genospecies (B. afzelii, B. garinii and B. valaisiana). Three samples from collection contained more than one genospecies. For other fifteen strains nucleotide sequences of flaB and ospC were determined. The phylogenetic analysis assigned our isolates to B. garinii (12) and B. afzelii (3) respectively, which were further subdivided into seven distinct ospC groups. The most related OspC types were G2, G4, G5 (B. garinii) and A3 and A8 (B. afzelii). |
