Popis | Combination of mass spectrometry and liquid separations is an essential analytical technique for protein research and the study of biomolecules in general. Driven by the need to identify, characterize, and quantify analytes at ever increasing sensitivity and speed, a range of analytical platforms and experimental strategies has emerged. Coupling column separation techniques to desorption mass spectrometry provides certain advantages over hyphenation with more common spray ionization techniques: separation is decoupled from mass analysis allowing re-analysis of selected peaks and avoiding a throughput mismatch where a too slow or a too fast mass spectrometer is connected on-line. Furthermore, the off-line arrangement allows employment of multiple detection techniques and on-target reactions. In the field of bioanalysis, matrix-assisted laser desorption/ionization (MALDI) offers dissimilar ionization efficiencies and reduction suppression effects compared to electrospray ionization (ESI). This lecture will provide an overview of deposition arrangements, ionization techniques and mass analyzers for both chromatographic and electromigration separations. Selected applications will cover both organic and inorganic analysis featuring the most prominent desorption ionization technique, MALDI as well as surface-assisted laser desorption/ionization (SALDI) and inductively coupled plasma (ICP) ionization. |
