Initial Insights into the Topology of Endoplasmic Reticulum localized PINs in Arabidopsis Thaliana

Popis | Auxin regulates plant growth and development through differential accumulation within tissues depending on the activity of PIN proteins that facilitate auxin efflux between cells. The plasma membrane PINs are involved in polar auxin transport (PAT), while the ER localized ones regulate intracellular auxin levels. The topology of plasma membrane (PM) localized ("long") PINs has recently been assessed. However, for the ER PINs this has not been done yet and we try to bridge this gap with our recent efforts. For the topology studies we have utilized GFP tagged PIN5 and PIN8 constructs, ectopically localizing on the PM of epidermis and cortex root cells. Here, we present experimental data showing the localization of PIN5 and PIN8 hydrophilic loops (HLs), using approaches based on pH-dependent quenching of fluorescent reporters, immuno-localization and protease protection techniques. The experiments were also guided by topology prediction software. |
