Eudragit® RL coated pellets with immobilized butyrylcholinesterase for rapid detection of cholinesterase inhibitors in water

Popis | Nerve agents (soman, sarin, tabun, VX), organophosphate (parathion, malathion) and carbamate (aldicarb) pesticides are commonly referred to as cholinesterase inhibitors (ChI), which are substances with very high toxicity. A fast, cheap, and sensitive sensor is needed to detect ChI as quickly as possible in order to carry out the appropriate treatment for those affected. Detection tubes (DTs) that use pellets with immobilized cholinesterase for colorimetric detection of the ChI presence, are just the type of detector that meets the above-mentioned requirements. Moreover, enzyme-based biosensors such as DTs have a detection limit between 0.005–0.01 mg•m-3, which is the lowest in comparison with more expensive and unportable detectors using GC-MS, FTIR or IMS. DTs are based on fast enzymatic Ellman reaction. However, it suffers from an indistinct color transition from white to yellow in the absence of ChI. In this study, an innovative carrier in the form of coated matrix pellets was prepared utilizing the advantages of the previous experiment. The pellet cores contained the unique sorbent – Neusilin® US2 and the semipermeable coating consisted mainly of Eudragit® RL. The main aim of the experiment was to evaluate reaction rate, sensitivity, and color transition intensity of prepared samples in comparison with previous types of carriers. |
