The interplay of δ-subunit and RNA polymerase in the pre-initiation state

Popis | In prokaryotes, RNA polymerase (RNAP) is a key element in the transcription of genetic information. In order to be efficient and tightly regulated, many microorganisms developed a wide array of transcription factors and auxiliary proteins. In some G+ bacteria, the RNA polymerase is associated with the ?-subunit. The roughly 20 kDa protein consists of 2 domains. The N-terminal which is ordered and facilitates binding to the core, whilst the long negatively charged C-terminal domain is disordered. Previous studies have shown both in in vivo and in vitro studies, that ?-subunit affects the transcription of many genes and its deletion can affect critical processes such as virulence and sporulation. Unfortunately, this step was never structurally characterized. Through the aid of cryoelectron microscopy, we try to uncover the possible interplay between the ?-subunit and the RNAP core, with a focus on the assembly of the pre-initiation complexes, including the ?-factors. |
