
Co-culture with CD40L, IL4 and IL21 expressing stromal cells induces robust CLL cell proliferation in vitro and in PDX co-transplantations

Popis | Several in vitro model systems have been developed to mimic CLL proliferation in lymph nodes typically utilizing CD40L-expressing stromal cells and addition of recombinant factors such as IL4 or IL21 or CpG. However, in such models, CLL proliferation is relatively low and variable. Here we aim to prepare a novel co-culture model based on mimicking T cell signals in vitro and in PDX. We prepared six cell lines by engineering by HS5 stromal cells with stable exypression of human CD40L, IL4. IL21, and their combinants, namely HS5-CD40L, HS5-IL4, HS5-IL21, HS5-CD40L-IL4, HS5-CD40LLOW-IL4, and HS5-CD40L-IL4.IL21 cells. HS5 cells were co.cultured with CLL cells for proliferation and gene expression studies, drug testing and co-transplated on 3D scaffolds into NSG mice. |
