Popis | The autophagic and endo-lysosomal pathways play a crucial role not only in degrading and recycling cellular waste, but in extracellular release and intracellular communication as well. Autophagy is a highly evolutionarily conserved pathway best known for its role in controlling protein and organelle turnover with a high impact on maintaining cellular homeostasis. Exosomes are a class of extracellular vesicles (EVs), originating from endosomes, released upon exocytosis. Exosomes and EVs in general play important roles in intracellular communication of normal and even tumor cells. The size and the cargo of exosomes are highly dependent on the cell of their origin. Additionally, exosomes can be formed after the fusion of autophagosomes and late endosomes, thus indicating the crosstalk between these two pathways. Inhibition of autophagy can significantly affect the secretion of extracellular vesicles, their quantity, but also their content. In this study, we inhibited the process of autophagy at different levels using commonly used and relatively new compounds, Subsequently, we evaluated changes in the proteome of EVs using LC-MS and western blotting. |
