Postreceptor effect of antidepressants on C6 glioma cells and NK lymphocytes

Popis | We studied antidepressant (AD) evoked cell signalling alterations in relationship to heterotrimeric GTP-binding (G) proteins, effector phospholipase C (PLC) and apoptosis. We used rat C6 glioma cell line, C6 glioma cells transfected (t) with pig thymus lymphocyte markers (MHCII, CD4 etc.)and natural killer (NK) lymphocytes. We analysed postreceptor effects of fluoxetine, FLU and sertraline, SER, specific serotonin reuptake inhibitors (SSR1). We estimated acute effects (20-24 h) and AD on Galfa subunits using ELISA and western immunoblotting with our antibodies. We demonstrated FLU effect on decreased level of Galfa q/11 in both C6 glioma and NK cells, in contrast to FLU induced changes of Galfas and Galfa(i1,2) in (t)C6 glioma cells. In C6 glioma cells, FLU induced Galga q/11 subunit translocation from membrane into cytosol and lower IP3 level, second messenger released by PLC and subsequent apoptotic events were observed. On the other hand SER effects on C6 glioma cells were linked to Galfys mainly without both IP3 changes and apoptosis. |
