Immunohistochemical analysis of some ECM components in the nerve outgrowth regenerating through the knitted tubes

Popis The synthesis of the extracellular matrix (ECM) is inevitable condition for the peripheral nerve regeneration. In our experiments, we investigated several components of the ECM (fibronectin, collagen IV, thrombospondin, chondroitin sulfate, and vitronectin) by the indirect immunohistochemical method in cryostat cross sections through nerve regenerates obtained from 6 adult female rats. The regenerate rejoined the proximal and distal stumps of the transected sciatic nerve that had been inserted into the guide prepared from the knitted vascular prosthesis. After 6 weeks from the transection, the regenerate consisted of a band of axons arranged in fascicles running along the longitudinal axis of the prosthesis. The immunostaining for fibronectin, collagen IV, thrombospondin, and chondroitin sulfate was present in the sections. The immunoreaction was found in the connective tissue at the surface of the regenerate as well as in the perineurium of nerve fascicles. No immunoreactivity for vitronectin was observed in the whole regenerate. Our immunohistochemical results suggest that the ECM produced in the knitted vascular prosthesis provides a milieu suitable for nerve regeneration.
