Popis Intracellular ROS production significantly regulates the fate of various cell types through redox interaction with signal transduction pathways (reviewed in Hensley et al. 2000). Thus, the determination of the actual intracellular ROS level in cells is crucial for understanding the ROS role in the key processes governing differentiation, cellular death and carcinogenesis. Intracellular ROS are usually produced by non-phagocytic NADPH oxidases and mitochondria (Kowaltowski et al. 2009). Thus, mitochondria mass and membrane potential are often determined together with intracellular ROS production. A wide spectrum of fluorescent redox-sensitive probes for the determination of intracellular ROS production have been developed and employed in numerous studies (Soh 2006, Dickinson et al. 2009). Both the high sensitivity and the reproducibility of results of are directly connected with the preservation of the oxidized/reduced fluorescent probe inside the cell.
