Using the high-resolution cytometry, we have established the positions of some genes, centromeres and chromosomes in nuclei of different cell types and their changes during basic cellular processes such as cell cycle, differentiation, repair of damage, e tc. We have also found that the changes in the gene location may correlate with gene silencing during differentiation, which strongly suggest that chromatin structure represents an important factor in the regulation of gene expression. in this project, t he local structure in the close neighbourhood of genes will be studied and correlation to gene expression will be investigated. We have already developed the spectral microscopy (SM) technique for these purposes which allows us to detect distances below 50 nm. As a model example, the globin locus will be analysed: the DNA probes will be prepared for the local control region (LCR) as well as for the beta-globin region (labelled with different fluorochrome) on the opposite sides of the locus and their dis
