Description | With help of high sensitive methods it was found that the vast majority of cervical dysplasias and carcinomas is causally connected to the human papillomavirus infection (HPV), especially to the high risk oncogen types (-16, -18, -31, -33, etc.). Human papillomaviruses in dysplasias and carcinomas of cervix transform the epithel among others by pRb inactivation by oncoprotein E7. Protein p16 is functionally connected to the cell cycle regulation through protein Rb. Protein Rb in its active form inhibits the p16 transcription. Resulting from the blocation of pRb function p16 value increases ineffectivelly; p16 does decrease the value of cyclin D/cdk4 complexes but it will not assert functionally itself because of pRb inactivation. It was found a higher expression of p16 in precanceroses and carcinomas. The aim of the study was to confirm the correlation between the p16 expression and dysplastic transformation of cervical epithelium and HPV 16, 18 incidence. We have used the histological samples for imunohistochemistry detection. We have demonstrated the existence of protein p16 in particular cell lines and in high grade lesions from the liquid based smears immunocytochemic and by immunoblotting and we have formulated these procedures metodically. |
